Tout sépare Lucas (Chad Michael Murray) et Nathan (James Lafferty). Lucas est un garçon calme et posé, fils unique d'une mère active et excellent joueur de basket-ball. Nathan est beaucoup plus prétentieux, star de son lycée et capitaine de l'équipe de basket-ball. Ils ont grandi dans la ville de Tree Hill en Caroline du Nord mais ne sont pas dans le même lycée. En fait, ils n'ont qu'un seul point commun, ils partagent le même père et sont donc demi-frères. Ils se retrouvent quand Lucas est transféré dans l'équipe de basket de Nathan. Commence alors une véritable compétition entre ces deux frères ennemis, d'abord sur le terrain, mais aussi pour le cœur de la jolie Peyton Sawyer (Hilarie Burton), la petite amie de Nathan dont Lucas tombe très vite amoureux.
